چکیده

ses. Enzymatic determinations demonstrated, however, that the enzymes of the glycolytic pathway (up to the formation of triose phosphates) were present and active at 25 and 37 C. At 42 C, the mutant did not grow with any of the carbon sources used. At both 37 and 42 C, the mutant grew perfectly well with hexoses if yeast extract was present. The metabolite required for growth was thiamine or, specifically, its thiazole moiety. If glucose was added to a culture growing in glycerol, at 37 C, growth was inhibited. This inhibition was relieved by the addition of thiamine or thiazole. Thus, at 37 C and only in the presence of hexoses, the mutant manifests a requirement for thiazole. This auxotrophy is absolute at 42 C. These data indicate that, in this mutant, some derivative of hexoses inhibits the synthesis of thiazole, and that this inhibition is also dependent on the temperature of incubation. The position in the bacterial chromosome of the genetic locus of this lesion (thz-) was determined by conjugation and found to coincide with the only thiamine (thi) locus so far reported.

In vitro Susceptibility Study of Salmonella typhimurium Mutant to Stress Factors

ABSTRACT         Background and Objective: In this study, we compared the susceptibility of Salmonella typhimurium phoP mutant and its parent to stress conditions that the pathogen may encounter in a host.         Methods: For this purpose, we used the phoP deletion mutant constructed in our previous study. In order t...

Isolation and Detection of Virulence factors of Salmonella typhimurium and Salmonella enteritidis

Plasmids play an important role in the virulence and antibiotic resistance of Salmonella. Salmonella carry plasmids having different size and number. Plasmid profile analysis of single isolates of Salmonella typhimurium and Salmonella enteritidis revealed the presence of 4- plasmid profiles. Salmonella serovars produce several Type III secretions including Sop-E and Sop –B. Soap E is detected b...

The multi-copper-ion oxidase CueO of Salmonella enterica serovar Typhimurium is required for systemic virulence.

Salmonella enterica serovar Typhimurium possesses a multi-copper-ion oxidase (multicopper oxidase), CueO (also known as CuiD), a periplasmic enzyme known to be required for resistance to copper ions. CueO from S. Typhimurium was expressed as a recombinant protein in Escherichia coli, and the purified protein exhibited a high cuprous oxidase activity. We have characterized an S. Typhimurium cueO...

ClpP is involved in the stress response and degradation of misfolded proteins in Salmonella enterica serovar Typhimurium.

Components of the ATP-dependent Clp protease complex are found in a wide range of prokaryotic cells and they are often expressed as part of the cellular stress response. To investigate the physiological role of the proteolytic subunit, ClpP, in Salmonella enterica serovar Typhimurium (S. typhimurium) an in-frame deletion of the clpP gene was constructed. Growth experiments revealed that clpP is...

Characterization of the Salmonella Isolates from Backyard Chickens in North of Iran, by Serotyping, Multiplex PCR and Antibiotic Resistance Analysis

The present study was designed to investigate the prevalence of Salmonella species, their molecular characterization and antibiotic resistance in backyard chickens. A total of 1125 samples were collected from backyard chickens in two consequent samplings. In the first part, samples were included of 820 poor recently hatched chicks, hatching residuals, egg shells in the nest floor, cloacal swabs...
